Development of a method for obtaining and using of tuberculous bacteriophage

Objective

The Project aim. Obtaining tuberculosis phage and its use in the development of methods for increasing effectiveness of tuberculosis control in animals.
Current status. Works on secretion of specific phages from epizootic Brucella cultures are currently carried out using inducing factor - penicillin. Spectrum of their lytic action had been studied. When studying a lytic spectrum it was established that the phage induced by penicillin can lyse strains of B.abortus, melitensis in such forms as S, - SR-and R unlike a referential TB phage forming negative colonies only on cultures of B.abortus and B. suis in S-form. Our experts carry out works on secretion of phage from objects of environment in TB unfavourable farms. At the present time our specialists develop the method for secretion TB phage which is similar to our previous method for secretion of Brucellosis phage. At the same time, their lytic activity to Mycobacterium will be studied.
The project’ influence on progress in this area. Preparation dosage form tuberculosis phage will allow its use as a high-preventive means.
The participants’ expertise. The participants’ expertise. Scientists involved in the project have extensive experience in the design, implementation and production of diagnostic, preventive drugs to protect animals against tuberculosis, as express methods for laboratory diagnosis of tuberculosis.
Expected results and their application. Based on the data from the research will assess the prospects of the use of phage-TV to develop methods for diagnosis and prevention of tuberculosis animals.
Meeting the ISTC goals and objectives. As the project participants are experts in the field of mass-destruction weapons and the project has exclusively peace orientation, it quite corresponds to ISTC objectives. Also, compliance to these purposes is reached at the cost of planned wide involvement of scientists from participating organizations in the world scientific community by presentation of the project data at international conferences and seminars.
Scope of activities. The following main works will be executed within the project:
- to conduct study, which includes search and isolation of tuberculosis phage (material for research is expected to use the environment - soil, manure, sewage, etc.);
- study of its morphological, serological and biological properties in vitro and in vivo;
- determining the possibility of using a dedicated tuberculosis phage for the diagnosis of tuberculosis;
- getting its experimental drug, as well as an integrated assessment of the prospects of the use of phage-TВ for the development of diagnostic methods and prevention of tuberculosis animals.
Role of Foreign Collaborators/Partners. Consultations, organization of business trips for preliminary interconnection with other partners, trainings, scientific exchange and coordination of joint actions, joint workshops (including working panel), meetings and consultations, technical report analysis, joint use of rare materials, samples and means, joint or parallel studies, discussion of obtained information, verification of results using self-contained methods and technology, joint publications and official registration of right fir intellectual property.
Technical approach and methodology. Under this Project will be conducted researches including search and isolation of TB phage (as a material will be used environment - soil, manure, sewage, etc.).
To achieve rapid detection of Mycobacterium tuberculosis (MBT) in clinical samples of pathological material will be used polymerase chain reaction (PCR);
Study of its morphological, serological and molecular-biological properties in vitro and in vivo. During this process will be used bacteriological, serological methods and biosamples on laboratory animals;
Determination of TB phage usability for diagnosis and prophylaxis of animal tuberculosis; for determination of TB phage usability will be used phagotyping method and phage titer increase reaction.
At prefinal stage will be performed in vivo experiments on guinea pigs. Based on obtained results of the above studies will be assessed the prospect of using TB phage to develop methods of diagnosis and prophylaxis of animal tuberculosis.
