Protection of Food Contamination by Novel Probiotic from Strains Lactobacillus and Propionibacterium

Project Status: 8 Project completed
Commencement Date: 01.08.2001
Duration in months: 34 months

Objective

The purpose of the project is the creation of new probiotics based on lactic-acid bacteria, effective at treatment of colibacteriosis and salmonellosis in chickens and porkers
Project topicality
The problem of effective treatment against acute intestinal infections in humans and animals is of great importance. The complexity of treatment of these diseases is based on the wide circulation of drug-resistance pathogens and the high frequency of disbacteriosis, caused by adverse factors, such as pollution of the environment, over use of antibiotics and other chemiotherapeutic compounds, influence of stress, and other factors. Therefore it is especially important to develop microbial preparations based on intestinal simbionts, which are ecologically pure and do not cause harmful by-effects. Known at the moment to be ineffective are preparations that cannot serve as alternative antibiotics. This testifies to the need for research in the creation of new effective probiotics for remedial-preventive purposes.
Expected results
From selected antagonistically active strains of lactic acid bacteria it is proposed to: 1) create a stable association at half-continuous and periodic cultivation, rendering bactericidal action against pathogens of colibacteriosis and salmonellosis, 2) allocate and identify antibiotic complexes, produced by individual Lactobacillus cultures and their associations, and 3) demonstrate antibacterial activity of these products on the basis of the suppression of growth of clinical strains of Salmonella and E.coli and their enzymic activity.
A consortium of lactic-acid bacteria and individual strains will be tested for their ability to prevent colibacteriosis and salmonellosis of chickens and porkers.
Meeting ISTC objectives
The project corresponds with the purposes of ISTC, as it enables scientists, formerly connected to weapons research, to utilize their expertise in peace activities, while supporting fundamental and applied research in environmental protection.
The work will be carried out by qualified experts: microbiologists, engaged in the isolation, selection and study of physiological and biochemical properties of lactic-acid and propionic bacteria, and who have experience in the creation of probiotics by directed action; chemists, whose expertise includes spectroscopy, stereochemistry and chromatography of biologically active substances; veterinary surgeons, working in the field of preventive maintenance and treatment of intestinal infections in agricultural animals and birds.
The work is designed for 2 years. During its fulfilment methods of selection of microorganisms, determination of antimicrobial and enzymic activity, vitamin-producing ability, light and electron microscopy, UV and IR spectrophotometry, different methods of chromatography will all be used.
This project is a joint project between the Oak Ridge National Laboratory (ORNL), the U.S. Department of Agriculture - Agricultural Research Service (USDA-ARS), ContiGroup Companies, Inc., and the Kazakh Institutes.
